Illumina iScan System

Overview

The Illumina iScan system is an array scanner that supports sensitive and accurate imaging of Illumina’s array based genetic analysis products.

Technology

Illumina bead-based microarray technology uses silica microbeads. As DNA fragments pass over the BeadChip, each probe binds to a complementary sequence in the sample DNA, stopping one base before the locus of interest. Allele specificity is conferred by a single base extension that incorporates one of four labeled nucleotides. When excited by a laser, the nucleotide label emits a signal. The intensity of that signal conveys information about the allelic ratio at that locus.

N-Counter Sprint Profiler

Overview

Digital examination of multiple RNA, DNA pathways in a single tube in an extraction free workflow.

Technology

Directly reads RNA by tagging it with capture probe and reporter probe creating a unique target probe complex. Excess probes are removed after hybridization. The complexes are then immobilized and aligned on the imaging surface. The sample is scanned by an automated fluorescence microscope and data analyzed with an analysis software

Digital Spatial Profiling

Overview

The GeoMx Digital Spatial Profiler (DSP) provides morphological context in spatial transcriptomics and spatial proteomics experiments from just one slide.

Technology

The tissue sections are stained using an mRNA or protein panel with DSP barcodes and immunofluorescent markers. Regions of interest are selected using GeoMx software, the barcodes are photocleaved and collected and quantified to create expression profiles.

Bioanalyzer

Overview

The Agilent 2100 Bioanalyzer system is an established automated electrophoresis solution for the sample quality control of biomolecules. The system integrates an instrument, data processing software, reagents, and a microfluidic chip specific for DNA, RNA, or protein analysis.

Technology

Conventional gel electrophoretic assays have been transferred to a chip format. DNA and protein samples are quantitated with the help of an upper marker or known standard while RNA samples are run alongside a ladder and quantitated. For total RNA assays, ribosomal ratio is determined which is an indicator of the integrity of the sample.

Key Features

  • The Agilent RNA 6000 Pico kit will accommodate up to 11 samples with concentration ranging from 50-5000 picograms per microliter for samples in water.
  • The Agilent Small RNA kit will accommodate up to 11 samples and will separate RNA fragments in the range of 6 to 150 nucleotides with a total RNA concentration 1-100 nanograms per microliter.”
  • The Agilent RNA 6000 Nano kit will take a maximum of 12 samples with the concentration range in 25 to 500 nanograms per microliter.
  • The Agilent DNA 1000 kit will accommodate up to 12 samples in the size range of 25 to 1000 bp and a concentration range of 0.1 to 50 nanograms per microliter.

Bioanalyzer Service

The service fee is $5 per sample. Please submit RNA samples in the concentration range of 15 to 500 ng/μl for use with the Nano chip. Bioanalyzer only orders with RNA samples below 15 ng/μl may use the Pico chip. An RNA Integrity Number (RIN) should be greater than 8.0.

Autoantigen Microarray

Overview

Autoantibodies are associated with various autoimmune diseases and dysregulation of the immune system. Profiling of autoantibodies against a broad range of autoantigens is a powerful tool for identification of biomarkers and for prediction and diagnosis of diseases.

Technology

A contact printing robot covalently attaches nanoliter volume of proteins to a nitrocellulose coated glass surface, the hybridization of autoantibodies present in the samples onto the coated antigens is detected by fluorescent tagged secondary antibodies.

Affymetrix

Overview

Affymetrix is a microarray platform that can evaluate the mRNA gene expression profile of thousands of genes in a single experiment.

Technology

The oligonucleotide probes on the array are hybridized with cRNA copies that are made invitro in the presence of biotinylated ribonucleotides. After hybridization, the chip is stained with streptavidin-phycoerythrin and scanned.

Quanterix Simoa HD-X Analyzer

Overview

Simoa (Single Molecular Array) HD-X analyzer at the IPC is one of the most sensitive automated bead-based immunoassay platforms with multiplexing and custom assay properties.

Technology

  • Paramagnetic beads coupled to capture antibodies are incubated with the sample analyte of interest and detection antibodies are added.
  • The beads are coupled to an immunocomplex consisting of the capture antibody-analyte-detection antibody.
  • The individual beads are trapped in femtoliter-sized wells along with a fluorogenic enzyme substrate, those that are bound by the immunocomplex are detected by fluorescence that is converted to a digital read.

Luminex

Overview

Our team has extensive experience in quantifying biomarkers in samples (serum, plasma, CSF and cell culture supernatant). We will work with you and help you find and perform appropriate commercially available assay or customize panels as per your requirements.

Technology

The Luminex® xMAP® technology is a bead-based immunoassay that allows for multiplex detection of up to 80 analytes simultaneously. Using dual laser flow-based detection, the first laser classifies the bead and identifies the analyte, the second laser detects the PE derived signal from the detected antibody bound to the analyte hence directly quantifying the marker of interest. Unlike ELISA assays that are dependent on colorimetric detection, Luminex is dependent on a fluorescence reporter system.
